Procedente de la exótica Estambul, en él viaja el detective Hércules Poirot, que repentinamente se topa con uno de los casos más desconcertantes de su carrera: en el compartimiento vecino ha sido asesinado Samuel E. Ratchett mientras dormía, pese a que ningún indicio trasluce un móvil concreto. Poirot aprovechará la situación para indagar entre los ocupantes del vagón, que a todas luces deberían ser los únicos posibles autores del crimen.Una víctima, doce sospechosos y una mente privilegiada en busca de la verdad. El Orient Express parte una vez más hacia los exóticos paisajes que jalonan su ruta de Estambul a Calais. Sin embargo, en esta ocasión el viaje no transcurrirá con la placidez de costumbre: el tren quedará bloqueado por la nieve y aparecerá el cadaver de un pasajero asesinado. ¿Quien puede ser el asesino de entre todos los viajeros? Después de haber resuelto un caso, Poirot viaja en el Expreso de Oriente, junto a un gran número de pasajeros. Ocurre entonces un asesinato dentro del tren y minutos después comienza a caer una tempestad de nieve que deja preso al Oriente Express. A partir de este momento todos los pasajeros pasan a ser sospechosos del asesinato. Mientras tanto Poirot descubre que la víctima era un asesino que, en un crimen conocido como "caso Armstrong".
The book is filled with entertaining and descriptive events that will leave readers anticipating more. Estambul, pleno invierno. Poirot decide tomar el Orient Express que en esta época suele hacer su recorrido prácticamente vacío. Pero aquel día, el tren va lleno y sólo gracias a una buena amiga consigue una litera en el coche-cama. A la mañana siguiente se despierta, descubre que una tormenta de nieve ha obligado a detener el tren y que un americano, llamado Ratcher, ha sido apuñalado salvajemente. Aparentemente nadie ha entrado ni ha salido del coche-cama. El asesino, sin duda, es alguno de los ocupantes entre los que se encuentra una altiva princesa rusa y una institutriz inglesa.
