There are many good bios of Jefferson, and these have been covered elsewhere on this forum (or maybe check the American Revolution forum). I am currently reading Thomas Jefferson: The Art of Power by Jon Meacham, which was just released this past November and is still on the nonfiction NYT bestseller list. It's shaping up to be a very good one-volume biography, although I think Meacham seems a little too starstruck of Jefferson at times - perhaps not enough coverage of his flaws (but I'm only half-way through).
Also check out Thomas Jefferson and the New Nation by Merrill D. Peterson and Thomas Jefferson: An Intimate History by Fawn Brodie.

Although it may say as much about the author as the subject, I also liked Christopher Hitchens's Thomas Jefferson, Author of America.
